چکیده

The new generation of centrifugal parallel fast analyzers, such as the “CentrifiChem,” is generally thought of primarily as enzyme analyzers, because of the several attractive characteristics they possess for such assays. With them, one can make an initial reading within several seconds of starting a reaction. They therefore can also be used to determine the concentration of selected compounds, by quantitating the rate of the reactions used; alternatively, endpointor equilibrium-reaction methods can be used. The determination of serum total protein provides an interesting case in point. Determination of serum total protein with the fast analyzer was first described by Hatcher and Anderson (1). However, they did not study many samples from patients nor did they discuss the characteristics and requirements of the kinetic approach. In this paper, we demonstrate the determination of serum total protein by kinetic analysis with the CentrifiChem Fast Analyzer.

Measurement of total protein and albumin in serum with a centrifugal analyzer.

In most clinical chemistry laboratories, mechanized procedures are essential for measurement of serum total protein and albumin. The biuret reaction is well established as the most practical approach to mechanized total protein measurements (1-4); albumin has posed a more difficult problem, with several procedures being proposed, some of which have involved salt fractionation (1-4), electrophor...

Stability in urea as a measure of cardiac lactate dehydrogenase on the centrifugal analyzer.

Kinetic methods are described for determination of total, heat-stable, and urea-stable lactate dehydrogenase (EC 1.1.1.27) activity on the centrifugal analyzer. In the urea-stable method we present, 2.6 molar urea is used to differentiate lactate dehydrogenase originatingfrom heart and liver. This urea concentration, greater than that used by most other investigators, better differentiates betwe...

Use of a Laser-equipped Centrifugal Analyzer for Kinetic Measurement of Serum lgG

Use of a visible laser in an Aminco “Rotochem” centrifugal analyzer enables light-scattering measurements to be made with this instrument. The modification is technically simple, inexpensive, and requires no fundamental changes in basic instrument design. The laser-modified analyzer has been applied to the measurement of lgG in serum. A two-point kinetic analysis is used to quantitate lgG from ...

New kinetic method for creatinine measurement now automated in the Cobas Fara centrifugal analyzer.

A new kinetic method to determine creatimne in serum by using an alkaline 3,5-dinitrobenzoate solution was recently reported (1). Interference from pseudo-creatinine chromogens, bilirubin, or triacylglycerols was negligible. The results obtained with this kinetic spectrophotometric assay, performed with an Eppendorf ACP 5040 analyzer, agreed well with the procedure of Jaff#{233}, as modified by...

An enzymic, reaction-rate assay for serum creatinine with a centrifugal analyzer.

We describe a procedure for specific, rapid, kinetic determination of creatinine, in which a manual coupled-enzyme micro-scale assay is adapted to a centrifugal analyzer. The creatinine reaction is ultimately linked to NADH utilization, which is measured by the absorbance change at 340 nm. This procedure requires 15 microL of serum and the standard curve is linear to a creatinine concentration ...
